Signosis’TFActivationProfilingArraysareusedformonitoringtheactivitiesofmultipleTFssimultaneously.Inthetechnology,aseriesofbiotin-labeledprobesaremadebasedontheconsensussequencesofTFDNA-bindingsites.Whentheprobemixturesincubatewithnuclearextracts,individualprobeswillfindtheircorrespondingTFsandformTF/probecomplexes,whichcanbeeasilyseparatedfromfreeprobesthroughasimplespincolumnpurification.Theboundprobesarethendetachedfromthecomplexesandanalyzedthroughhybridizationwithaplate,whicheachwellisspecificallypre-coatedwithcomplementarysequencesoftheprobes.
