Mitogen-activatedproteinkinase(MAPK)signalingpathwaysmediateimportanteukaryotictranscriptionalresponsestoextracellularsignalsthatbeginatthecellsurface. WhiledifferentexternalstimulimayactivatedistinctMAPKsignalingpathways,manyMAPKpathwayslaterconvergeintheactivationofETSdomaintranscriptionfactorELK1. ELK1isamajornuclearsubstrateforMAPKs,JNK,p38,andERKpathways,wherephosphorylationofELK1bythesekinasesresultsinaconformationalchangeofELK1andtriggersitsDNAbindingactivity.
SignosishasestablishedanELK-TADluciferasereporterstablecellline. ThiscelllineexpressesafusionproteinlinkingtheDNAtransactivationdomain(TAD)ofELKtoGal4. AstheTADisphosphorylated,thisGal4fusionproteinwilldriveluciferaseexpressioncontrolledbyGal4-UAS. ThiscelllinecanactasareportersystemtomonitorELK1-dependenttranscriptionalactivityandMAPKsignaling,astriggeredbystimulitreatment,enforcedgeneexpressionandgeneknockdown.
