TheseactivatedSmadscomplexwithSmad4(co-Smad),translocatefromcytoplasmintonucleusandbindtotargetpromoterregiontoregulategenetranscriptions. DysfunctioninTGFβpathwayleadstoimmunosuppressionandangiogenesis,whichcanmakecancermoreinvasive.

Onceactivated,theTFtranslocatestothenucleusandofteninteractswithrelevantco-factorstodrivegeneexpression.
